Immunohistochemical analysis of secreted from muscle stage larvae (SML)-2. SML-2 protein was localised in formalin-fixed, paraffin embedded Trichinella spiralis-infected rat tissue collected 28 days p.i. (A–C) Thin sections were probed with anti-SML-2 (A and B) or anti-SML-2 pre-bleed control sera (C) and detected with horseradish peroxidase-conjugated anti-IgG and 3,3′-diaminobenzidine substrate. Sections were counterstained with Mayer’s haematoxylin. The black arrows show areas of intense staining within the nematode. Localisation of SML-2 in adults and uterine NBLs was also examined. Trichinella spiralis adults were isolated 6 days p.i., formalin-fixed and paraffin embedded. Thin sections were cut to poly-lysine coated slides and probed with anti-SML-2 (D) or SML-2 (E) pre-bleed control sera. Black arrows indicate areas of cross reactivity either in the stichocytes of the adults or small vesicles in the uterine NBLs (D) or equivalent anatomical sections probed with pre-bleed sera (E).
