IBOP induces translocation of 3xHAhTPα from the ER to the Golgi. TPα-HEK cells were treated with vehicle or IBOP (100 nM, 3 h). A, B: Cell lysates were fractionated on a discontinous density gradient and resolved by SDS-PAGE. Fractions were identified with marker proteins for Golgi (golgin-97; green arrows) and ER (calnexin; blue arrows). Western blots are representative of three independent experiments. C: HAhTP (red staining) and Golgi (green staining) were visualized in intact cells by immunofluoresence microscopy. Perinuclear staining for TPα was observed in untreated cells, consistent with localization to the ER. Colocalization of HAhTPα and Golgi is in yellow (arrows). Images are from a representative experiment that was repeated with similar results.
