Fifty picomoles of each substrate was incubated with 2 mU BgaC or BgaA in a 50-μl reaction mixture (50 mM NaPO4 [pH 6.5], 10 mM MgCl2, 45 mM β-mercaptoethanol) for 20 h at 30°C. Release of the terminal galactose from substrates was analyzed by HPLC or MALDI-TOF. The sugar chains 025, 028, 042, 043, and 044 were purchased from Takara (Japan). The sugar chain NA2 was from Sigma.
