Analysis of UL138 transcripts. (A) Total RNA isolated from FIXwt-infected (MOI = 2) MRC5 cells over a time course was analyzed by Northern blotting using a radiolabeled antisense UL138-specific probe. To determine the kinetics of UL138 expression, cells were treated with CHX or PAA and RNA was isolated at the times indicated. (B) Poly(A)⁺ purified RNA isolated from infected MRC5 cells at 24 hpi was analyzed by Northern blotting using radiolabeled antisense probes specific to the ULb′ ORFs indicated or a sense probe specific to UL138.
