ICP27 hypomethylation affects its cellular localization. (A) HeLa cells were infected with wild-type (WT) HSV-1 KOS, ΔRGG, or the indicated ICP27 arginine-to-lysine viral mutants. At 5 and 8 h after infection, the cells were fixed and immunostained with anti-ICP27 monoclonal antibody. (B) HeLa cells were either mock infected or infected with WT HSV-1 KOS. The general methylation inhibitor AdOx was added 2 h after infection to the indicated samples. At the times indicated, cells were fixed and immunostained with anti-ICP27 or anti-hnRNPA2 monoclonal antibodies.
