Receptor-stimulated Cdc42 activity is dependent on ERM activity. Jurkat cells were transfected with Raichu-Cdc42, either alone or with the indicated ezrin (VSV-G-tagged) construct, and allowed to adhere to anti-CD3 (UCHT1) (A) or anti-β1 integrin (12G10) (B) antibody-coated coverslips for 30 min at 37°C before fixation. Exogenous ezrin expression was detected by staining (following cell fixation and permeabilization) with a Cy5-conjugated anti-VSV-G IgG. (C) Bar charts represent the cumulative data of three experiments (n = 10 cells error bars ± standard errors of the means). We compared each treatment group to the controls and other groups by one-way analysis of variance (ANOVA). If the ANOVA indicated a significant difference between groups, a pairwise multiple comparison of all means and post hoc testing using Tukey's method were employed to determine significant differences between groups and controls.
