Binding of wt and mutant L4-100K to eIF4G and viral late mRNAs. (A) A549 cells were infected with H5pg4100 and H5pm4151 viruses at a multiplicity of 10 FFU per cell. At 48 h after infection, total cell lysates were prepared and subjected to immunoprecipitation (IP) with L4-100K antibody, eIF4G antibody, or Ab412 (recognizing methylated arginines). The immunocomplexes were separated by SDS-10% PAGE and analyzed by immunoblotting using MAb 6B10. (B) RNA was purified from immunoprecipitates of total cell lysates obtained from infected cells grown in the absence (AdOx−) or presence (AdOx+) of AdOx. RNA was reverse transcribed and subjected to quantitative real-time PCR as described in the text. L5 fiber and TL RNA levels are shown relative to the wt values.
