Recombinant Purified ΔLTPG Binds the Lipophilic Probe TNS.

Recombinant LTPG (ΔLTPG) was engineered to resemble the mature processed protein by omitting both the N-terminal signal sequence and GPI-anchor domain and then purified following overexpression in E. coli. ΔLTPG, incubated with increasing concentrations of TNS, exhibited saturation binding defined by a Kd of 12.7 ± 2.2 μM (circles). No saturation binding was observed (squares) if the protein was denatured by boiling in a solution of 6 M urea and 200 mM DTT.
