FIGURE 2. — Txnl1 is associated with the 19 S regulatory complex. A, HeLa cell extracts were separated by nondenaturing PAGE and analyzed by immunoblotting (IB) using antibodies against a 20 S subunit (α4), a 19 S subunit (Rpn2), and Txnl1. As a control, a purified sample of His₆-Txnl1 was also subjected to nondenaturing PAGE and Coomassie-stained. His₆-tagged Rpn11 (B) or His₆-tagged Csn5 (C)(lane 1) was precipitated with glutathione-Sepharose beads loaded with GST (lane 2) and GST-Txnl1 (lane 3). Precipitated material was separated by SDS-PAGE and analyzed by immunoblotting with an antibody to the His₆ tag.
