TABLE 3.
Maximal mineralization of [14C]formaldehyde by the wild type and isogenic P. putida mutantsa
| Strain | Level of 14CO2 (1,000 cpm) from:
|
|
|---|---|---|
| HCOH | HCOOH | |
| Parental strain | 10.5 ± 0.3 | 207 ± 10 |
| PP0328 mutant | 8.1 ± 0.2 | 175 ± 5 |
| PP3970 mutant | 6.2 ± 0.1 | 218 ± 5 |
| PP0328/PP3970 mutant | 6.1 ± 0.4 | 162 ± 7 |
| PP0489 mutant | 5.1 ± 0.2 | 142 ± 3 |
| PP0492 mutant | 6.4 ± 0.3 | 113 ± 2 |
| PP2184 mutant | 6.8 ± 0.1 | 133 ± 1 |
| PP0489/PP2184 mutant | 2.7 ± 0.1 | 103 ± 1 |
The indicated strains were grown in LB medium overnight and then diluted 100-fold in the same medium supplemented with 0.5 μCi formaldehyde or 5 μCi formate. Evolved 14CO2 was trapped in a solution (5 M KOH) placed in a vessel fixed to the bottom of the incubation flask. Values are those after 24 h of incubation. Other conditions are as given in Materials and Methods.
