Human articular chondrocytes express both 15-LOX-1 and 15-LOX-2. (a) Chondrocytes were isolated from OA knee cartilage and maintained as monolayer culture for 7 to 10 days. Total RNA was prepared, reverse transcribed into cDNA, and processed for PCR using specific primers for 15-LOX-1, 15-LOX-2, and GAPDH. PCR products were resolved on a 1.8% agarose gel and stained with ethidium bromide. C-RT and C-PCR are negative controls for the reverse transcription and PCR reaction, respectively.
