Autologus CD8+T cells with DC showed higher cytolytic activity against BEC-2 target cells at a high effector-to-target ratio than against Bamb-2 target cells. Autologous CD8+T cells were co-cultured with M1-, M4- or TNF-α-primed DC loaded with EBV-derived peptide, expanded with 100 U/ml IL-2, and on day 10, re-stimulated with BEC-2 or Bamb-2 target cells at various effector-to-target ratios. After overnight culture, the IFN-γ concentration in the supernatant was measured by ELISA (A), and specific lysis was measured by 51Cr release assay (B).
