A. Hippocampal neurons were pre-treated with the nSMase2 inhibitor GW4869 (10 μM) or the serine palmitoyltransferase inhibitor ISP-1 (10 μM) then exposed to TNFα for 2 min. GW4869, but not ISP-1 prevented TNFα from increasing the clustering of NR1S896 into GM1+ lipid rafts. Data are mean ± S.D. from a minimum 21 dendrites/condition derived from 3 separate experiments. B. TNFα (50 ng/ml) induced clustering of NR1S896 into GM1+ domains in wt/wt mouse hippocampal cultures was slightly reduced in cultures from wt/fro mice and was not different from vehicle controls in neurons cultured from fro/fro mice. ### = p < 0.001 compared with vehicle and *** = p < 0.001 compared with TNFα or +/+.
