shRNA inhibition of NFAT5 transcriptional and TNF promoter activities. mTAL cells were incubated in the absence or presence of 1.2 mM CaCl₂ for 6 h after transfection with pTonE_Luc (A) or a TNF-luciferase promoter construct in combination with constructs to inhibit NFAT5 (U6-N5 ex8) or NFAT1 (U6-N1 ex4; B), as indicated.
