Limited proteolysis of wild-type and Y341A TyrRS to probe for structural changes. (A) Additional bands (boxed) were observed with Y341A TyrRS using either plasmin (top) or elastase (bottom) for digestion. (B) The cleavage sites generated by plasmin (magenta) and elastase (cyan) are marked on the structure. Additional cleavage sites were generated for Y341A relative to wild-type TyrRS, and those additional sites are located on or near helix α14.
