2×10⁵ purified CD8⁺ T cells were cultured with allogeneic purified CD11c⁺ DC for 3 days prior to the addition of [H³] thymidine. Proliferation was measured by the incorporation of [H³] thymidine above that of syngeneic cultures. (A) B6 T cells with BALB/c DC or BALB/c T cells with B6 DC are inhibited by the addition of anti-CD40L to the cultures (B) CD40L^−/− T cells respond equally as well as wildtype. (C) CD40L^−/− DC are poorer presenters to BALB/c T cells (D) CD40^−/− T cells proliferate to a lesser extent (**p=0.003, Mann Whitney U Test). Mean + SEM are shown.
