Replication kinetics of wild-type HIV-1 and mutants F130W and T58S/F130W in MT-4 cells. MT-4 cells (6 × 106) were infected at a multiplicity of infection of 0.0001 TCID50 per cell. Infections were monitored by measuring RT activity (a) and p24 production (b). Cell lysates collected when the p24 levels reached the highest values (day 4 for wild-type HIV-1, day 6 for mutant T58S/F130W, and day 9 for mutant F130W) were analyzed by Western blot (c – f). Immunoblots were probed with monoclonal antibodies against p24 (c), IN (d), the RNase H domain of the RT (e), and the RT heterodimer (f). Samples were normalized using a β-actin-specific antibody.
