ADP inhibits the reopening of WT-CFTR by MgPPi. (A) WT channels were first opened by ATP. Once current reached steady state, a direct switch of the solution to MgPPi (dead time, ∼30 ms) resulted in a rapid decrease of the current, followed by an increase of the current. In the same patch, the effect of MgPPi was dramatically reduced when applied together with 5 mM ADP. (B) Summary of data presented in A. Steady-state current induced by MgPPi or MgPPi plus ADP was normalized to original ATP-dependent current. IMgPPi/IATP = 0.53 ± 0.036 (n = 5); IMgPPi-ADP/IATP = 0.065 ± 0.014 (n = 5; P < 0.01).
