Asparagine (Asn) deamidation in proteins is highly correlated with Asn–Gly (glycine) sequences. In this spontaneous, non-enzymatic reaction, the amide N of the Asn side chain attacks the peptide carbonyl of the following Gly to form a transient cyclic succinimide. The latter is hydrolyzed by water to yield both D- and L-aspartate (Asp) in both α and β linkages. A β-linked aspartate is known as isoaspartate (isoAsp).
