(A) Western blot analysis, (B & C) The ratio of normalized CE cell phospho-CREB (against the actin internal standard) levels over that averaged from the control CE cells of the same experiment (Y-axis) was shown as a function of VIP concentration (B) and time of VIP-pretreatment (C). For B: The ratios were (mean±sem [p values in control vs VIP-pretreted]): 1±0.085, 1.43±0.08 (p=0.03), 1.97±0.18 9 (p<0.0001), and 1.46±0.28 (p=0.04) in 0, 10−10, 10−8, and 10−6 M VIP-treated CE cells in 10, 2, 7, and 6 corneoscleral explants, respectively. Data were combined from three separate experiments. The difference among various groups of corneoscleral explants was significant (P = 0.006, ANOVA).
