ASB2 ubiquitin ligase activity triggers degradation of FLNa and -b in myeloid leukemia cells. PLB985/MT-FLAG-ASB2wt and PLB985/MT-FLAG-ASB2LA cells were left untreated (−) or were treated (+) with 100 μm ZnSO4 (Zn) for 8 h. A, cytosolic extracts (15 μg) were separated by SDS-PAGE and immunoblotted for ASB2, FLNa, FLNb, Talin 1, and non-muscle myosin heavy chain IIA (NM Myosin IIA). B, 150-μg aliquots of cytosolic extracts of PLB985/MT-FLAG-ASB2wt and PLB985/MT-FLAG-ASB2LA cells cultured with 100 μm ZnSO4 for 8 h were loaded on an SDS, 7% polyacrylamide gel. Proteins were revealed by colloidal Coomassie Blue staining. Molecular mass marker proteins were run in parallel.
