A Cultured cardiac myocytes were treated with 10⁻⁷ mol/L ET-1 in the presence or absence of 400 nmol/L TSA, as indicated. Nuclear extracts were incubated with antibody to YY1, followed by immunoprecipitation and Western blot analysis with an antibody to HDAC2. Results are representative of three independent experiments. B. Cardiac myocytes were transfected with -198 hBNP LUC or control pcDNA3 plasmid. Cells were pretreated with 400 nmol/L TSA for one hour and then treated with 10⁻⁷mol/L ET-1 for an additional hour, as indicated. Cells were collected and the DNA-IP assay was performed as described in Methods. Results are representative of three independent experiments.
