(A) GM2AP extracts DDHPE from liposomes. (B) Preincubation of GM2AP with GM2 micelles inhibits the extraction of DDHPE. 10 μM GM2AP was preincubated for 20 min with varying concentrations of GM2 micelles, which ranged from 0 to 15 μM. The GM2AP-GM2 mixture was then allowed to incubate with POPC:DDHPE (2:1) vesicles for 20 min, followed by ultracentrifugation at room temperature. For both A and B, the change in the DDHPE concentration in the supernatant was determined from fluorescence measurements as described in the Materials and Methods section. Data points represent the average value from three separate experiments with data points representing the standard deviation in A and error bars showing the standard deviation in B. All experiments were performed in 50 mM NaOAc, 100 mM KCl, pH 4.8 with 100 μM final lipid concentration.
