In the presence of SCD-1, hepatic saturated fatty acids (SFA) are converted into monounsaturated fatty acids, which increase SREBP-1c maturation, lipogenesis, and TG/CE synthesis. In the absence of SCD-1 (SCD-1−/−), the inability to desaturate hepatic SFA leads to an inability to upregulate de novo lipogenesis via SREBP-1c and storage as TG. Therefore, β-oxidation and uncoupling (not shown) increase to decrease TG and VLDL.
