Mutations in the −144/−135 Sp1 element do not limit the CREB or NF-κB-mediated regulation of HKα2 proximal promoter activity. pGL3–0.18mHKα2 or pGL3–0.18mHKα2Δ−144/−135 and the Renilla luciferase expression plasmid pRL-TK were cotransfected in the presence of the expression vector for NF-κB p65 or the constitutively active CREB1 mutant CREB-VP16. Forty-eight hours after transfection, cell lysates were prepared and firefly and Renilla luciferase activities in lysates of the cells were assayed.
