Shown are representative examples of COS-1 cells expressing the YFP-tagged wild-type secretin receptor and the 14 TM IV cysteine-replacement mutants. All except for G241C, W242C, and P245C mutants show significant YFP fluorescence on the cell surface. All images were acquired under similar settings and were representative of at least three independent experiments.
