Comparison of IL-15-induced WT vs GzmB−/− cre NK cell granzyme B and C mRNA levels. Flow-sorted NK cells (≥95% NKp46+CD3) were cultured in K10 medium with recombinant murine IL-15 (100 ng/ml), and total RNA was harvested for qRT-PCR at various times during activation. The data shown are fold change (mean ± SEM) of granzyme B and granzyme C mRNA in IL-15-activated NK cells, compared with fresh (day 0) NK cells, in three independent experiments. The comparative Ct method was used and granzyme B and C mRNA abundance was normalized to β-actin.
