Schematic depiction of Adeno-hTERT-E1A genome construction. Transcription factor-binding sites in the hTERT core promoter fragment (−371 to +27) are indicated in our core hTERT promoter (step 1), which was cloned upstream of the viral E1A gene in the pGL3 vector. Next, the hTERT-E1A-polyA cassette was excised with MluI and BamHI, blunt ended, and then ligated into the EcoRI site of an intermediate pShuttle vector (step 2) for the ultimate viral genome-cloning step (step 3), using the Adeno-X system genome.
