AG1478 represses miR-21 expression. H3255 lung adenocarcinoma cells, characterized by a high expression of miR-21 and EGFR mutation, were serum starved for 24 h and then were grown in either the presence or absence of AG1478 (2 μM or 10 μM) for 2 h with or without following exposure to 20 ng/mL EGF for 15 min. (A) The effect of AG1478 on p-EGFR and p-Akt expression. β-actin was a loading control. (B) MiR-21 expression levels analyzed by qRT-PCR after the AG1478 treatments (2 μM or 10 μM) with or without EGF ligand stimulation.
