L. major glycoconjugate molecules do not affect the expression of costimulatory molecules but influence IL-12 production by DCs. (A) BMDCs were infected with CFSE-labeled WT, lpg2 AB and lpg2-deficient (lpg2) L. major stationary-phase promastigotes (BMDC/parasite ratio, 1:10), and the rate of infection was determined after 24 h by gating on infected cells (CD11c+ CFSE+ cells). The expression of MHC class II, CD40, CD80, and CD86 on infected (CFSE+) or on total (CD11c+) DCs was determined by flow cytometry. Data are presented as percent MFI of gated infected cells (B). Some uninfected and infected BMDCs were stimulated with various concentrations of anti-CD40 and IFN-γ for 24 h, and the culture supernatant fluids were collected and assayed for IL-12p40 by ELISA (C).
