Time course and magnitude of amplificatory shift is not dependent on Cl− driving force. Results from a typical cell clamped with a perforated gramicidin patch to maintain intracellular Cl− levels. Extracellular Cl− was altered to 5 mM (top, Cm traces), 140 mM (middle, Cm traces) and 1 mM (bottom, Cm traces). Voltage protocol is shown at bottom. After step jumps to either 100 or −100 mV from 0 mV, a ramp was voltage delivered to gauge the shift of Vpkcm. Shifts were −47.8 to −40.9, −50 to −41.5, and −54 to −46 for 5, 140, and 1 mM Cl−, respectively.
