Schematic of the optical setup used for single molecule spectroscopy. 488 nm and 568 nm lasers were overlapped and focused on a submicrometer fluidic channel using a high numerical aperture microscope objective. Multicolor fluorescence emission was collected, split, filtered and focused on 50 mm optical fibers. Avalanche photodiodes were used for signal transduction and amplification.
