TnI mutations have little effect on the concentration of free troponin required for 50% inhibition of actin-activated S1 ATPase activity. Actin-activated ATPases for WT and mutants at 0.05 M ionic strength are shown with increasing concentrations of troponin in the absence of Ca2+. Initial velocities (μM ATP/s/μM S1) are shown for WT and D191H (A), R193H (B), R146G (C) and R146W (D). All rates were corrected for the ATPase rate of S1 in the absence of actin (0.06 μM ATP/s/μM S1). Data are presented as mean values ± SD.
