Sterol analysis of macrophages treated with ketoconazole and econazole

RAW 264.7 Mϕs were treated with ketoconazole (1 μm), econazole (10 μm), or the respective carrier (DMSO) for 16 h. Sterols were saponified by incubation of cell pellets in MeOH, 60% KOH (2:1), extracted and analyzed by gas chromatography. Ergosterol was added as an internal standard, and samples were corrected by both ergosterol for extraction and protein levels for input.
