Motility of MCF-10A cells engendered by PKCα or S165D-α6-tubulin is eliminated by treatment with nocodazole. Cells were transfected with 4 μg of plasmid DNA encoding PKCα, S165D-α6-tubulin, or the vector control, as described under “Materials and Methods.” After 48 h, cells were plated onto 10-well slides, and, following incubation at 37 °C and 5% CO₂ overnight, the cells were treated (t = 0) with 5 μm nocodazole (or 0.1% (v/v) DMSO) and analyzed for motility after 8 h. Each measurement is the average of triplicate measurements ± S.D., and the results are representative of three independent experiments.
