Molecular beacon assay for Ape1. (A) An oligonucleotide containing a tetrahydrofuran (THF) abasic site mimic was used in the Ape1 molecular beacon substrate. The top strand was labeled on the 5’ end with a 6-carboxyfluorescein (FAM) fluorophore, the bottom on the 3’end with a dabsyl (DAB) quench. (B) Upon cleavage by Ape1, the fluorophore dissociates from the dabsyl quench, causing an increase in fluorescence. (C) An approximate 6-fold increase in fluorescence is observed upon DNA cleavage by Ape1.
