Iron-binding constants. Double reciprocal plot displaying initial rate of NADH-dependent acetyl-CoA and acetaldehyde dehydrogenase reaction as a function of Fe²⁺ concentration. Fe²⁺ was removed from partially purified EhADH2 with phenanthroline. The concentration of Fe²⁺ present in the standard activity assay buffer was then varied between 10 and 500 µM, and the initial rate of reaction was measured.
