CsA-resistance mutations alleviate a lethal mutation at the C-terminal domain of NS5B. (A) Transient replication of Rep1b, P540A, and the derivatives. One μg of in vitro transcribed replicon RNAs was electroporated into Huh-7.5 cells, and total RNA was isolated 7 h, 4 days, and 10 days after electroporation. The ratio of HCV RNA to the GAPDH RNA was plotted for each mutant at each time point. The mock-transfected cells were electroporated with buffer.
