Purified splenic B cells from VH12 transgenic mice (VH12-Tg) and negative littermate controls (VH12-NL) were stimulated with indicated antibodies for 15 min at 37°C and immunoblotted for pAkt (at S473). Anti-IgM indicates the antibody without biotin and is used at 50 μg/ml. Anti-μ~bio and anti-CD19~bio indicate biotin conjugated antibodies and are respectively used at 0.5 and 2.0 μg/ml. 10 μg/ml avidin is used to cross-link IgM and CD19. This is a representative of at least 3 independent experiments.
