14D degradation by propane-grown cultures of Graphium sp. Cultures of Graphium sp. were initially grown on PDB and then incubated with propane as described in Materials and Methods. The PDB-grown, propane-induced mycelia were then transferred to glass bottles (700 ml) containing MSM (50 ml). The bottles were sealed, and if required, acetylene (2% [vol/vol] gas phase) was added. The reaction was initiated by the addition of 14D (10 μmol), and at the indicated times, the concentration of 14D was determined by GC as described in Materials and Methods. The figure shows the time course for 14D consumption for incubations conducted in the presence (▵) and absence (▴) of acetylene. The data presented show the means and standard errors for three replicate incubations.
