Silencing the expression of the proteins that form the DLP has a differential effect on viral translation. MA104 cells in 48-well plates were transfected with the indicated siRNA as described in Materials and Methods. At 48 h posttransfection, the cells were infected with RRV at an MOI of 3 (A, B, and C) or with rotavirus strains with RRV, YM, and SA11 (D), and at 7 hpi the cells were radiolabeled for 60 min with 25 μCi/ml of Easy-tag Express-35S and then lysed. The labeled proteins were resolved by SDS-10% PAGE (A, B, and D) or by SDS-7% PAGE (B) and detected by autoradiography (left panels) or by immunoblot analysis (right panels) using either a mixture of antibodies to VP1 and VP4 (B) or a polyclonal antirotavirus antibody (C).
