PAR3 overexpression enhances thrombin-elicited ATP release and inositol phosphate formation in A549 cells. A, cells were transfected with the indicated amount of cDNA. Forty-eight hours post transfection, cells were preincubated with ebselen and βγ-metATP as in Fig. 1 and incubated for 5 min with 30 nm thrombin or vehicle. B and C, cells transfected with 100 ng of PAR3 cDNA were stimulated for 5 min (B) or 20 min (C) with the indicated concentration of thrombin, and ATP release and inositol phosphate formation were measured as described in Fig. 1, C and D, respectively. The data represent the mean ± S.E. of at least three independent experiments performed in quadruplicate.
