(A) Wild-type (WT) or ΔsetA L. pneumophila strains were cultured in broth and equivalent A₆₀₀ units were harvested at different stages of growth. Lysates were prepared and immunoprobed with affinity-purified antibodies directed against recombinant SetA. DotG immunoblot is a control for equivalent loading conditions. Cell density is indicated in A₆₀₀ units at the top of the blot. Arrow indicates the band corresponding to SetA. (B) Primary bone-marrow derived macrophages from A/J mice and D. discoideum were infected with the indicated strains. Viable counts were determined as described in Experimental Procedures. The means and standard deviations are shown for each time point; experiments were done in triplicate.
