(A) Human fibrosarcoma cells (HT1080) suspended in serum-free Dulbecco's modified Eagle's medium containing 0.1% bovine serum albumin were seeded (1 × 104 cells/well) on 8 μm pore Transwell filters coated with Matrigel (50 mg/well) in the absence or presence of increasing concentrations of inhibitor 2 (0–10 μM). The lower well was filled with medium containing 5% fetal bovine serum, as a chemo-attractant. After 18 h incubation, the cells that migrated to the underside of the filters were fixed, stained and photographed. Five different fields were counted and the average number of migrating cells/filter was calculated. Each value represents the mean ± standard deviation of triplicate wells.
