E6* does not interact with either Akt or Dlg in vitro. 293 cells were transiently transfected with constructs expressing HA-tagged Akt or Dlg. After cells were harvested, 10% of each whole-cell extract (WCE) was retained as a control for input levels of the expressed proteins, and the remainder was used for pull-downs with GST, GST-E6* (GST 18E6*), or GST-E6 (GST 18E6) bound to glutathione resin. After the resin was washed, bound proteins were eluted and visualized by SDS-PAGE and Western blotting with anti-HA antibodies. The membranes were subsequently stained with Ponceau Red to determine input levels of the GST fusion proteins.
