Altered GEF activities of the Epac2-F435G and Epac2-F435W mutants. Activation of Rap1 by Epac2-F435G (A) or Epac2-F435W (C) in the absence or presence of various concentrations of cAMP was monitored by following the time-dependent decrease in fluorescence intensity as a measure of the rate of exchange of Mant-GDP by GDP. Dependence of Epac2-F435G (B) or Epac2-F435W (D) GEF activity (kGEF) on cAMP concentration is shown.
