Ribonuclear foci in subsynaptic nuclei of muscle fibers. (a) In sections of DM1 skeletal muscle, FISH demonstrates foci of CUGexp RNA (red) in subsynaptic nuclei (DAPI, blue). Endplates are shown by staining with α–BTX (green). (b) Immunofluorescence for MBNL1 (red) reveals MBNL1 foci in subsynaptic myonuclei (DAPI, blue; α–BTX, green), suggesting sequestration of MBNL1 by CUGexp RNA. (c) In HSALR mice, foci of CUGexp RNA (red) are abundant in extrajunctional myonuclei (arrows), but sparse or absent in myonuclei (DAPI, blue) subadjacent to the endplate (α–BTX, green).
