Aryl 1-indanylketone 1 inhibits the chemotactic activity of CypA but not CypB. Activated CD4+ T cells were purified using MACS separation after overnight stimulation of spleen mononuclear cells with ConA. Chemotaxis assays were conducted using a 48-well modified Boyden chamber. RANTES was used as a positive control for cell migration. CypA and CypB were used at 100 ng/ml and 200 ng/ml, respectively. Compound 1 was used at 2 μM. Data show mean (± SE) chemotactic indices for each group with n=6 wells per group. A chemotactic index was generated for each well by dividing the number of cells migrating within each well by the average number of cells migrating to medium alone.
