LPS pre-incubation protects against H2O2-induced barrier loss independent of NOS2. MTE cells from C57Bl6/J (A) or NOS2-/- mice (B) were pre-treated with LPS (10 μg/mL) for 24 hrs, and subsequently exposed to H2O2 (500 μM) for 3 hrs during which TER was monitored. The NOS2 specific inhibitor 1400W (100 μM) was added 30 min prior to H2O2 exposure. (C) RT-PCR analysis of LPS-induced expression of NOS2 mRNA. Data were expressed relative to initial TER for each well.
