Western blot analysis of SDS-PAGE of wild type and mutant HA-hPAT1 protein under nonreducing (−DTT) and reducing (+DTT) conditions. Lysates of oocytes expressing either wild type or mutant transporter were alkylated and separated under reducing (100 mm DTT) or nonreducing (0 mm DTT) conditions on a 7.5% SDS gel (10 × 10 cm), followed by immunodetection with the mouse anti-HA tag antibody.
